2013年7月28日讯 /生物谷BIOON/ --由National Institute of Allergy and Infectious Diseases (NIAID)

A monoclonal antibody developed by researchers at the National Institute of Allergy and Infectious Diseases (NIAID) could lay the groundwork for effective therapies against norovirus, which has in recent years infected school campuses, cruise ships, hospitals and nursing homes.

In an upcoming study in the Journal of Virology, the NIAID scientists provide the first proof-of-concept data showing that a monoclonal antibody can neutralize human norovirus, nicknamed the winter vomiting bug. Currently, there are no vaccines or specific antiviral therapies available to treat norovirus.

"We initiated this work because there is presently no virus-specific treatment or vaccine to control the norovirus illness," said Kim Green, a researcher on the study, in a release. "Our working hypothesis was that a highly specific norovirus antibody that binds to the outer surface of the virus particle might prevent the ability of the virus to infect susceptible host cells."

To make the antibody, the researchers isolated genes from chimpanzee immune cells encoding norovirus-specific antibodies, then converted them into full-length immunoglobulin molecules compatible with the human body. Two of the antibodies were effective at combating norovirus infection in chimpanzees, according to the study.

In the U.S. alone, norovirus causes about 20 million cases of diarrhea and vomiting annually, according to the Centers for Disease Control and Prevention. In their study, the researchers said the antibody could be useful for treating high-risk populations, including infants and young children, the elderly, and patients with weak immune systems. For most people, symptoms of norovirus last an average of one or two days, but for vulnerable populations, norovirus can be life-threatening.
